Escherichia coli host strain for genome integrated Fab FTN2 expression.
H<dFTN2> originates from an Escherichia coli HMS174(DE3) strain and contains a Fab FTN2 expression cassette on the attTn7 site of the genome. FTN2 is a humanized Fab that neutralizes tumor necrosis factor α. For FTN2 translocation into the periplasmic space, the light chain (LC) and the heavy chain (HC) of the Fab were fused to the DsbA signal sequence (dsbA_SS), which directs the transport of the fused protein across the plasma membrane into the periplasm using the SRP pathway. doi: 10.1002/BIOT.201800637.
